Naldebain is indicated for the relief of moderate to severe acute postsurgical pain.  It has been used across many different types of operations to relieve pain.  The medicine is a sterile oil solution which is suitable for intramuscular injection and from a single injection it can provide pain relief for up to seven days.  Naldebain is a synthetic nalbuphine prodrug whose potency is similar to morphine, but without morphine’s risks of addiction and respiratory depression, which makes Naldebain a safer long-acting postoperative analgesic.

Naldebain has been proven to have significant post-operative analgesic efficacy. Naldebain patients reported lower pain scores than patients taking a placebo, whether measured 48 hours or 7 days after surgery.

Naldebain reduced the dosage of the rescue medication by 64% in post-operative 0-48 hours and by 43% in 48-168 hours.
The most common treatment emergent adverse events were injection site reaction (27.5%), pyrexia (16.5%),  dizziness (6.4%), vomiting (2.8%), nausea (1.8%) and somnolence (0.9%)[1].

The parent compound of Naldebain is nalbuphine which has a dual agonist/antagonist mechanism of action. Nalbuphine is effective at minimizing common opioid AEs such as pruritus, the severity of nausea and vomiting by rebalancing opioid μ and κ neuronal activity[2]. Naldebain also carries less risk of respiratory depression due to its ceiling effect.
